Another in Bond.  The spy who is glamorous is defined by James Bond and he is defined by Goldfinger.   

The 39 steps by hitchcock as defining the everyman getting caught in a spy web.

but for pure govt agent spy it seems to be Dr. No, IMO.

I don’t think “genre-defining” means first, it means the best or most platonically ideal example. Although often the they can be the same.  It’s a movie you recommend to someone and say “this is what genre X means.” 

Batman Begins: Comic book movies with no sense of humor about themselves.

Little Mermaid: The Mouse conquers all media; modern animated movies built around songs. 

The Shining: Psychological horror.

The Maltese Falcon: Film noir. 

Fast Times at Ridgemont High: Coming of age

Airport may have been first but I’d think Poseidon Adventure was the catalyst for disaster movies… Earthquake, Towering Inferno, Ishtar, Hindenburg, Meteor, several Bee movies.

Stagecoach is a good choice for Classic Western but I’d also submit The Searchers.

some are hard like Westerns and WWII movies because there are so many of them and have been made for decades in different styles.  I want The Wild Bunch to be one but then how do you describe it vs. westerns before it as a genre.

as always with these threads to each his own and some people have different opinions on defining a movie genre.  It kind of has to be extremely unique when it came out.

 

I would lobby for a subgenre of Westerns and would nominate Shane (1953) starring Alan Ladd in the Lone *Anti-Hero category with Unforgiven (1992) as a modern contender in that subgenre as well. It's a tricky category within Westerns as there have been quite a few solo lead movies in Westerns both before and after the George Steven's directed the highly nominated film that debuted in the 50's but I think it is well known enough among cinephiles to hold the lead in that category.

 

*I went with anti-hero as I am rather fond of morally gray characters. This wasn't the Lone Ranger.

Several of these movies RE defined genres.  Unforgiven, for example.  Same for John Wick.  It's an action movie with lotsa gun play, but totally changed action movies with lotsa gun play.

North by Northwest by Hitchcock starring Cary Grant defined and established this genre. Even the Broccolis acknowledge this. They'll tell you that Fleming's Bond doesn't get made without NxNW.
